The time after cocaine administration for which cocaine and its metabolites can be detected in plasma and urine depends on the sensitivity of the utilized assay method. Advise patients that cocaine hydrochloride and its metabolites in cocaine hydrochloride may be detected in plasma for up to one week after administration. Cocaine hydrochloride and its metabolites may be detected in urine toxicology screening for longer than one week after administration [see Warnings and Precautions ( 5.4)]. Cocaine has been described in literature to be primarily metabolized and inactivated by non-enzymatic ester hydrolysis and hepatic carboxylesterase, and also by plasma cholinesterase, hepatic carboxylesterase, and CYP3A4. The pharmacokinetics of NUMBRINO in patients with reduced plasma cholinesterase activity has not been studied. The testing liquid in the ampoule will start out golden yellow. Once you’ve mixed the sample in, wait approximately 10 seconds for a color change to begin. This test kit will react to levamisole, acetaminophen/paracetamol, phenacetin, aspirin, Vitamin C, ephedrine (HCL), lidocaine, benzocaine, and procaine, with various color changes to indicate which ingredient was detected. Cocaine is a stimulant, and it increases your heart rate and your rate of breathing, as well as your blood pressure and body temperature. This can make you feel hyper-stimulated, excited, and even anxious, paranoid, or irritable. Unfortunately, cocaine is damaging to the tissues of the nose (if it’s sniffed) and the respiratory system (if it’s smoked). It can also cause abscesses and infectious diseases if it’s injected. Taking it greatly increases your risk of heart problems, seizures, and death. Cocaine Purity Test Kits: If your sample does contain cocaine, this type of drug test kit will let you know how pure the cocaine is. Even high-quality cocaine isn’t going to be 100% pure, but performing this test kit will give you a good indication of whether or not you’re getting ripped off with a substance that contains very little actual cocaine.

With a large number of drug testing kits to choose from, knowing what products you can trust can be challenging. There are a lot of cocaine test kits on the market in 2023, but certain manufacturers and products are absolute standouts in terms of accuracy, price, and ease of use. In a Phase 3 study, patients received a mean dose of 126 mg (80 to 160 mg, N=259) of cocaine hydrochloride using NUMBRINO nasal solution 4% and a mean dose of 319 mg (200 to 400 mg, N=259) of cocaine hydrochloride using NUMBRINO nasal solution 10% as a single application.Cocaine hydrochloride is associated with increases in heart rate. The mean placebo corrected change from baseline heart rate (90% two-sided upper confidence interval) are 12 (14) bpm and 20 (22) bpm for the 4% and 10%, respectively.

Each test comes with an ampoule and a spatula for putting a sample inside.Reproductive effects:- On an acute basis, cocaine can prolong the time to reach orgasm in men and women. Delusions may respond to neuroleptics (phenothiazine and butyrophenone) but these agents also may increase the chance of seizures; benzodiazepines may be useful in reducing anxiety.

Advise patients of the signs and symptoms of hypersensitivity reactions and to seek immediate medical attention should they occur [see Warnings and Precautions ( 5.5)]. The most common adverse reactions reported with cocaine hydrochloride 4% are included in Table 1 (preexisting nasal conditions are not included). There were two patients treated with cocaine hydrochloride nasal solution 4% who withdrew due to an adverse reaction. One patient developed anxiety and systolic hypertension and one patient developed intermittent paroxysmal tachycardia. Both patients developed symptoms within 10 minutes of nasal pledget application. Three patients treated with cocaine hydrochloride nasal solution 10% required premature removal of pledgets due to nausea and diastolic hypertension; mild intermittent paroxysmal hypertension and paroxysmal tachycardia; and vasovagal syncope with bradycardia. Cocaine is taken from the coca plant, which grows in Peru, Argentina, Colombia, and other parts of South America. Chewing coca leaves for their stimulant and pain relief effects was the most common form of ingestion until cocaine was properly extracted and discovered in 1860. Epinephrine, Phenylephrine: There have been reports of myocardial ischemia, myocardial infarction, and ventricular arrhythmias with concomitant use during nasal surgery. Avoid use of additional vasoconstrictor agents with cocaine hydrochloride. If concomitant use is unavoidable, prolonged vital sign and ECG monitoring may be required. ( 5.3, 7.2) When it comes to getting fake drugs, it’s easy for many people to have an “it only happens to other people” attitude and assume that because they’ve been ok in the past, there’s no need to test in the future. The authorities and numerous harm reduction organizations would disagree, however, based on the statistics. Over the past several years, police have confiscated an increasing amount of cocaine that’s been cut with substances that are often dangerous and sometimes quite deadly. One spray delivers 130µl of solution (containing 13mg of Cocaine). Therefore, a maximal dose of 1.5mg/Kg of cocaine (approx. 1ml of 10% solution), is equivalent to approximately 8-9 sprays for a 70Kg adult, and this dose must not be exceeded. Cocaine hydrochloride may also be applied as a topical solution with a cotton swab. It is effective for numbing mucous membranes in other parts of the body, especially the mouth and throat.

Cocaine applied to the mucous membranes may be detectable for as long as three hours after its application. A true biological half-life for cocaine is difficult to ascertain, as serum concentrations reflect a dynamic balance between the absorption and degradation of cocaine, i.e. the concentration of cholinesterase in the blood can vary between individuals. Genetic abnormalities of plasma cholinesterase (e.g., patients who are heterozygous or homozygous for atypical plasma cholinesterase gene), disease conditions such as malignant tumors, severe liver or kidney disease, decompensated heart disease, infections, burns, anemia, peptic ulcer, or myxedema or other physiological states such as pregnancy may lead to reduced plasma cholinesterase activity. Patients with reduced plasma cholinesterase (pseudocholinesterase) activity may have reduced clearance and increased exposure of plasma cocaine after administration of cocaine hydrochloride. Cocaine hydrochloride causes numbness and vasoconstriction (narrowed blood vessels), which can reduce bleeding and swelling on the operating area. There are no available data on the use of cocaine hydrochloride in pregnant women to identify a drug-associated risk of major birth defects, miscarriage or adverse maternal or fetal outcomes. Adverse maternal and fetal/neonatal outcomes have been seen in women with chronic cocaine abuse during pregnancy ( see Data).

Cocaine hydrochloride is largely (90%) metabolised by cholinesterase, thus those patients taking cholinesterase inhibitors such as Ecothiopate eye drops for the treatment of glaucoma, or neostigmine for the treatment of Myasthenia Gravis, or those patients with hereditary Pseudocholinesterase deficiency, should not be administered cocaine hydrochloride. If these patients are given cocaine hydrochloride, higher blood levels result, with a greater risk of drug toxicity.
